Common steps for making a TMA block from formalin-fixed and paraffin-embedded (FFPE) tissue samples
Step 1:
Mark the coring area(s) on a H&E or other stained slide under a microscope.

Step 2:
Align the marked coring area(s) back to the donor block.

Step 2a:
Based on the alignment, mark the coring area(s) on the donor block.

Step 3:
Mark the borders of a blank recipient TMA block.

Step 4:
Insert and secure the recipient TMA block in Quickarrayer block holder and turn on the arrayer LED. Set the position slide near the first column as the starting line.

Step 5:
Create a hole through the arrayer guide plate on the recipient TMA block with an appropriate punching pen.

Step 6a:
Punch a marked area vertically on the donor block to obtain a tissue core with an appropriate coring pen. The illumination of our small or medium-sized light box greatly helps the identification of the coring area, especially for tiny and thin samples.
Step 6b:
Coring through a guide plate with larger hole can prevent damage to a donor block, especially for large tissue cores and harden or cold FFPE tissue blocks.
Step 6c:
Coring can be performed at selected angles or directions to obtain area of interest on a donor block. The illumination of our small or medium-sized light box greatly helps the identification of the coring area, especially for a tiny and thin samples.



Step 6d:
For heavily cored or damaged donor blocks, tissue fragments can also be collected by our coring pen. The illumination of our small or medium-sized light box greatly helps this process.
Step 6e:
Our coring pen can also collect loose FFPE tissue fragments on a smooth and hard surface (glass, metal, plastic….).
Step 6f:
After the collection of tissue fragments in 6d, 6e, press and tap the coring pen head against the hard surface to compact the fragments inside the coring head tube before Step 7.



Step 7:
Eject the tissue core inside the coring pen collected in Step 6 into the previously made hole (Step 5).

Step 7a:
A tissue core is ejected in the defined position of a recipient TMA block.

Step 8:
Completion of a TMA block. With our Quickarrayer, repeat Step 1 to 7, TMA blocks with different core sizes and layouts can be easily and precisely created.
